Effect of iron loading on human monocytes in vitro. Human monocytes were isolated from peripheral blood and incubated in six-well plates. The cells were incubated with iron (200 μM) or control overnight and then stimulated for 6 h with lipopolysaccharide (LPS; 150 ng/ml) or medium. Lipopolysaccharide alone induced Il6 mRNA (A) and Tnfα mRNA (B). Iron loading significantly increased mRNA levels in response to LPS (two-way ANOVA with Bonferroni post hoc tests; P values adjusted for all possible comparisons; n = 3 replicates/condition).
